Mechanism of cellular uptake and cytotoxicity of paclitaxel loaded lipid nanocapsules in breast cancer cells

Lipid nanocapsules (LNCs) have proven their efficacy in delivering different drugs to various cancers, but no studies yet described uptake mechanisms, paclitaxel (PTX) delivery or resulting cytotoxicity towards breast cancer cells. Herein, we report results concerning cellular of LNCs and PTX-loaded (LNCs-PTX) on the three cell lines MCF-7, MDA-MB-231 MDA-MB-468. LNCs-PTX sizes 50 ± 2 nm, 90 3 nm 120 4 were developed by phase inversion method. Fluorescence microscopy flow cytometry used observe fluorescently labeled was measured using HPLC analyses samples. These revealed a higher MDA-MB-468 cells than other two lines. Moreover, free PTX exhibited similar pattern toxicity each line, appeared be more sensitive lines, as evaluated MTT assay proliferation based upon [3H]thymidine incorporation. Studies with inhibitors endocytosis indicate that is mainly via Cdc42/GRAF-dependent well macropinocytosis, whereas dynamin-dependent processes are not required.
